Diet medicine Acomplia Found to Up Risk of Anxiety and Depression in Heart Study

Diet medicine rimonabant (Acomplia / Zimulti) produced disappointing results in slowing progression of coronary artery disease in patients and it significantly raised the risk of anxiety and depression, according to a report in the April 2 issue of the Journal of the American Medical Association. The results of the trial called STRADIVARIUS, released a day early to coincide with a presentation at the annual conference of the American College of Cardiology, seem certain to increase questions as to whether this once promising diet medicine will ever be approved in the U.S. because of side effects.

More than 40 percent of patients who took rimonabant in the STRADIVARIUS trial developed psychiatric problems and one committed suicide, the researchers reported.

While Acomplia is sold in European Union countries and some other nations, Sanofi Aventis pulled back its U.S. application last year after an FDA advisory panel recommended against approval of rimonabant because of concerns over psychiatric risks.

The STRADIVARIUS study, which involved 839 abdominally obese patients with coronary disease and metabolic risk factors at 112 centers, sought to determine whether rimonabant might help slow progression of atherosclerotic disease, often commonly known as “hardening” of the arteries.

The researchers reported that the study failed to meet its main goal of reducing the Percent Atheroma Volume (PAV), a measure of plaque in a cross-section of the arteries, although it did reduce the Total Atheroma Volume (TAV), a measurement of total plaque in the coronary artery.

“Because the current study failed to achieve a statistically significant effect for the primary efficacy measure, additional studies will be required to further define the role of rimonabant in the treatment of abdominally obese patients with coronary disease and metabolic risk factors,” the researchers concluded.

But more attention seems certain to be focused on this latest effort to assess the extent to which Acomplia carries with it an increased risk of psychiatric side effects.

The researchers in the STRADIVARIUS study included patients with a history of such problems, and reported that 43.4 percent of patients who got Acomplia suffered psychiatric side effects such as depression and anxiety compared to 28.4 percent of patients who were in the control group.

Dr. John Rumsfeld of the Denver Veterans Affairs Medical Center and Dr. Brahmajee Nallamothu of the University of Michigan, in a commentary in the medical journal, observed that while the promise of Acomplia remains, “it certainly has faded.

“The potential benefits are offset by the very real risks of depression and anxiety,” they added.

Fast-Track Your Fat Loss!
10-minute workouts that slim you down and tone all your trouble spots

The Expert: Chris Freytag, contributing fitness editor and model for our story, created this workout based on her forth-coming book and new DVD, both titled Shortcuts to Big Weight Loss.

“No time” is no longer an excuse for skipping a workout. Just ask Prevention’s contributing fitness editor, Chris Freytag, who developed this supereffective program that will shape you up on your schedule. As a working mother of three who drives car pools, corrects homework, and travels up to 2 weeks each month, Chris knows a thing–or five–about living a hectic lifestyle and keeping trim. Her secret weapon: do-anywhere 10-minute routines that burn calories and tone every trouble spot. Even better, the latest research shows that they’re more effective than traditional half-hour workouts. Almost instantly, you’ll have more energy. Within 2 weeks, you’ll feel stronger and look firmer. In 6 weeks, you could lose up to 10 pounds!

Put It All Together
What you need
A pair of supportive walking shoes, a set of 5- to 8-pound dumbbells, a chair or low table, and an exercise mat or a soft, carpeted area.

How to do it
Choose your goal below (lose weight, get toned, or boost energy and get healthier). Each one has a target number of sessions you need to complete each week. You’ll mix and match our various 10-minute cardio workouts and our 10-minute firming routines weekly to achieve your desired results.

When to do it
Ideally, aim to fit in 2 or 3 sessions throughout each day–but if you can’t, simply adjust your workouts over the week. For example, if you have time for only 1 or 2 workouts one day, double up another day when you have more time. Just try to get all your sessions in by the end of the week.

Here’s how a typical day could pan out:

6:50-7:00 am
Energy-Boost Walk

12:20-12:30 pm
Calorie-Burn Walk

5:15-5:25 pm
Arm Sculptor

8:50-9:00 pm
Belly Flattener

Do these: To Lose Weight
5 Energy-Boosters
6 Calorie-Burners
4 Toning Walks
2 Butt & Thigh Firmers
2 Arm Sculptors
2 Belly Flatteners
Total: 21 sessions each week

Do these: To Get Toned
3 Energy-Boosters
2 Calorie-Burner
6 Toning Walks
3 Butt & Thigh Firmers
3 Arm Sculptors
3 Belly Flatteners
Total: 20 sessions each week

Do these: To Boost Energy And Get Healthier
4 Energy-Boosters
4 Calorie-Burners
4 Toning Walks
1 Butt & Thigh Firmer
1 Arm Sculptor
1 Belly Flattener

Total: 15 sessions each week

10-Minute Workouts: Walking Workouts
Each of these three 10-minute routines revs up your calorie burn; we’ve tailored them even further for variety and to help you reach your specific goal. All three maximize every minute of your workout time by getting your heart rate up quickly and keeping you in the optimal fat-burning zone throughout the 10-minute routine. Intensity levels are based on a 1-to-10 scale, with 1 being how you’d feel standing still and 10 how you’d feel sprinting. And because the workout is short, you’ll have more energy to give it your all the entire time and burn more fat in the process.

To Firm Even Quicker
We have 1-minute toners that you can do intermittently almost anywhere to shape up in less time, or use them on days when you can’t fit in a full 10-minute workout.

For Faster Results
One Minute Toner: Upper Body
At home or in the office: Do standing push-ups. Place hands on a desk or counter and bend elbows, lowering chest, then push back up.

10-Minute Workout: Strength Routines
These sculpting workouts do double duty. Each includes 1-minute cardio bursts so you can rev your calorie burn while you tone.

Butt & Thigh Firmer

Warm up by marching in place while swinging your arms at your sides for 30 seconds. Rest 10 to 20 seconds between exercises. Repeat routine twice.

ACTIVITY
REPETITIONS

Clock Work
4 clocks with each leg

Cardio Burst: Side-to-Side Shuffle
1 minute

One-Legged Lunges
8 times with each leg

Cardio Burst: Side-to-Side Shuffle
1 minute

Monster Squats
8 times with each leg
1. Clock Work Stand with feet together. Step left foot forward (12 o’clock position) 2 to 3 feet, bend knees, and lower into a lunge, keeping front knee behind toes, as shown. Don’t lean forward. Press into left foot and stand back up, feet together. Step left foot out to side (9 o’clock position), feet hip-width apart. Bend knees and hips and sit back into a squat, knees behind toes, chest lifted. Stand back up, feet together. Step left foot behind you (6 o’clock position), lower into a lunge, then stand back up. Repeat with right leg stepping to 3 o’clock position for squat

2. One-Legged Lunges Place left foot on chair or table behind you so leg is extended and you’re balancing on right leg. Bend right knee, lowering into lunge, front knee behind toes. Press into right foot standing back up.

3. Monster Squats Stand with feet wider than shoulder-width apart, toes pointing out slightly, arms relaxed at sides. Sit back into a squat, keeping knees behind toes. As you stand up, raise left knee out to side, placing foot back on floor as you lower into next squat, lifting right leg.
Cardio Burst: Side-to-Side Shuffle
Sit back into a squat, with arms bent in front of you, elbows by sides. Shuffle 10 to 15 feet to the right, moving right foot then left foot. Return shuffling to left.
10-Minute Workout: Arm Sculptor
Warm up by marching in place while “swimming” your arms as if you were doing the breaststroke for 30 seconds. Rest 10 to 20 seconds between exercises. Repeat routine twice.

ACTIVITY
REPETITIONS

Supported Curls
15 times with each arm

Cardio Burst:
Mountain Climbers
1 minute

Side Plank Push-Ups
8 times on each side

Cardio Burst: Mountain Climbers
1 minute

T-Stand Rows with Kickbacks
15 times, balancing on left leg first time through and right leg second time through
1. Supported Curls Sit in a chair with feet a few inches apart. Holding a dumbbell in each hand, lean forward from hips and rest elbows on thighs, palms up. Bend left elbow, curling dumbbell toward shoulder. Keep upper arm still. Hold for a second, then lower and repeat with right arm. Continue alternating arms.

2. Side Plank Push-Ups Start on knees, hands beneath shoulders, and body in line from head to knees. Bend elbows out to sides, lowering chest almost to floor. Straighten arms, pushing back up, then raise left arm overhead, rolling body to left to form a side plank. Lower and repeat on right.

3. T-Stand Rows with Kickbacks Stand with feet together, a dumbbell in each hand. Hinge forward at hips and raise right leg behind you, arms hanging beneath shoulders, palms in. Tighten glutes and abs to stay balanced. Bend elbows toward ceiling and squeeze shoulder blades together, pulling dumbbells toward rib cage. Keeping upper arms still, press dumbbells back and straighten arms. Hold for a second, then reverse direction.
Cardio Burst: Mountain Climbers
Get in push-up position with hands on seat of chair. bring right foot forward 10 to 12 inches, knee bent. jump quickly, switching feet as if you were climbing a mountain.

For Faster Results: Upper Arms
At the store: Bend elbows, curling grocery bags toward shoulders. Hold for a second, then slowly lower.
1. Crisscrosses Lie faceup with knees above hips, calves parallel to floor, hands behind head. Contract abs, raising head and shoulders off floor as you extend right leg. Twist to left, bringing right elbow and left knee toward each other. Don’t pull on your neck; the work should come from your abs. Hold for a second, then switch sides, twisting to right. (That’s 1 rep.)
2. Full-Body Roll-Ups Lie faceup with legs extended, arms overhead. Contract transverse abdominis (the deep ab muscle that runs between your hip bones) as you inhale and raise arms up toward ceiling. Exhale as you tuck chin and roll up, pulling navel toward spine. Curl forward with arms extending in front of you. Inhale as you stay rounded. Slowly and with control, reverse directions, uncurling body as you exhale and lower to floor, one vertebra at a time. Keep feet on floor the entire time.
Cardio Burst: Bob ‘n’ Weave
Stand with feet shoulder-width apart, arms bent. Squat and dip head and torso to right as you stand back up, like you’re ducking under something. Repeat to left.

For Faster Results: Belly & Butt
In the car or standing in line: Alternate between contracting your abs and glutes, holding for 30 seconds each.

Why 10-Minute Workouts Are Better
Here are the benefits you’ll enjoy if you follow Chris’s plan:

You’ll lose 30% more fat
Women who performed 10-minute bouts of exercise throughout the day lost nearly a third more weight than those who exercised in one 30- to 40-minute chunk 3 times a week, reports a University of Pittsburgh study.

You’ll cut your heart attack risk in half
All it takes is accumulating an hour of walking a week. (You can do that!) Other studies show that multiple short workouts are more effective at lowering triglycerides (a type of blood fat that increases your heart attack risk) and raising “good” HDL cholesterol than a continuous 30-minute bout.

You’ll shrink your belly in less time
Women ages 31 to 57 who squeezed in three 10-minute walks most days of the week lost nearly twice as many inches off their waistlines as those who did single 30-minute sessions, according to British research.

You’ll get fit faster
Men and women who did two 15-minute workouts 4 days a week improved their cardiovascular endurance twice as much as those who worked out in a single 30-minute routine 4 days a week. Miniworkouts deliver such powerful results because you’re more likely to do them, say experts.

Obesity, a problem of our society

Obesity is a major health concern; and obesity is more prevalent than ever before in today’s modern society. Our lifestyles these days are just not conducive to healthy weight management, and we all spend too much time sitting behind a desk, and not enough time getting regular exercise. Many of us are so busy that we don’t even have the time to eat right. The toll of this unhealthy lifestyle is excess fat that builds up over the years, and eventually becomes a major health threat. Obesity has been proven to cause increased cancer prevalence’s, higher risks for heart disease, high cholesterol and high blood pressure. The statistics say that if you are obese, you will not live as long. It can be difficult to lose weight. Anyone who has tried to diet knows how difficult and frustrating the process can be. Your body is accustomed to certain caloric intakes, and your body wants to regulate that intake at a steady level. If you try to cut down your caloric consumption, your body will react, and you will feel constant hunger.(pill rimonabant)
Diet pills have been available for some time; but most of these drugs either didn’t work, or they did work, but the side effects and dangers were so severe, that it simply wasn’t worth continuing. Thankfully, that has now changed. The release of Acomplia (pill rimonabant) now provides obese people with a safe and effective medical aid for weight loss.
Clinical studies have proven Acomplia to be the safest and most effective weight loss drug ever developed. Acomplia will help you lose weight, safely.

A pill that helps you lose weight and quit smoking? That was amazing enough to capture headlines last week. But scientists say the experimental drug might be even more versatile, providing a new tool to help people stop abusing drugs and alcohol, too. It’s called Pill Rimonabant, or Acomplia (search), and last week researchers reported it could help people not only lose weight but keep it off for two years. That burnished the drug’s reputation after two studies in March, which suggested it could fight both obesity and smoking, two of humanity’s biggest killers. The French pharmaceutical firm Sanofi-Aventis plans to seek federal approval for Pill Rimonabant next year. But the drug’s benefits may go beyond just smokers and obese people, researchers say. “I think it’s going to have a big impact on the treatment of addiction,” said Dr. Charles O’Brien, an addiction expert at the University of Pennsylvania and the Philadelphia Veterans Affairs Medical Center. Animal studies suggest Pill Rimonabant can block the effects of marijuana and fight relapse in alcohol and cocaine abuse, he said. Once it is approved for treating obesity or smoking, “we’ll be free to study it in these other areas and I’ll try to get my hands on it as quickly as possible,” O’Brien said. He’s not alone in his enthusiasm. The National Institute on Alcohol Abuse and Alcoholism is interested in seeing whether Pill Rimonabant (search) can help treat heavy drinkers, said Dr. George Kunos of the institute. No human test results for Pill Rimonabant in alcohol abuse have yet been published, he said. But researchers at the National Institute on Drug Abuse reported in 2001 that a single dose of the drug could block the effects of smoked marijuana in people, not just animals. That suggests the drug could be useful in treating marijuana dependence (search), said Marilyn Huestis, principal investigator of the study. The institute is now pursuing follow-up research, said Huestis, acting chief for chemistry and drug metabolism research at NIDA.Pill Rimonabant’s versatility traces back to its effects on the brain’s reward system, circuitry that tells you to keep on doing something. Basically, it appears to help break the connection between an activity like smoking and the rewarding feeling it causes in the brain. The body has its own marijuana-like substances called endocannabinoids, and they activate certain brain cells that in turn can lead to stimulation of the brain’s reward system. Pleasurable things like drinking alcohol are thought to activate a feeling of reward by acting through the endocannabinoid (search) system.”We think that the (endocannabinoid) system is overactivated by chronic smoking, or perhaps even excessive overeating,” said Dr. Robert Anthenelli of the University of Cincinnati College of Medicine and the Cincinnati Veterans Affairs Medical Center. He’s on the advisory board of Sanofi-Aventis.Pill Rimonabant blocks the effect of the natural endocannabinoids by keeping them from latching onto the brain cells they normally stimulate, he said. In smokers, for example, that seems to restore the natural balance of the brain reward circuitry, he said.

Based on the data available from various clinical trials, Rimonabant (Acomplia, Zimulti, buy rimonabant pills) is an extremely safe medication with minor side effects such as nausea, mood swings, anxiety and dizziness.Rimonabant is recommended to be taken along with regular exercise and a diet regime for optimum results. Patients with a Body Mass Index (BMI) greater than or equal to 30kg/m2 can benefit from taking a daily pill of Rimonabant of 20mg along with proper exercise and diet.Various trials conducted have shown remarkable results making Rimonabant (Acomplia, Zimulti) one of the most sought after anti-obesity medications worldwide.

Rimonabant is also known as wonder drug as it treats obesity and smoking cessation simultaneously.Rimonabant (brand name Acomplia, buy rimonabant pill) is an anorectic anti-obesity drug. It is a CB1 cannabinoid receptor antagonist. Its main avenue of effect is reduction in appetite. Rimonabant is currently being sold in the United Kingdom by Sanofi-Aventis and in Denmark by Sanofi-Synthelabo under the trade name Acomplia. It is believed, although not confirmed, that if the drug is approved in the United States, it will be sold under the trade name Zimulti.(buy rimonabant pill)

Rimonabant is the first selective CB1 receptor blocker to be approved for use anywhere in the world. In Europe, it is indicated for use in conjunction with diet and exercise for patients with a body mass index greater than 30 kg/m2, or patients wih a BMI greater than 27 kg/m2 with associated risk factors, such as type 2 diabetes or dyslipidaemia. In the UK it has been available since the end of July, 2006.Rimonabant may also be found to be effective in assisting some smokers to quit smoking. Sanofi-Aventis is currently conducting studies to determine the possible value of rimonabant in smoking-cessation therapy.

One-stop shopping fans take note: a new pill that claims to help people stop smoking and lose weight at the same time is in its final stages of testing. It could be on the market by 2006. The new drug is called Rimonabant Acomplia, and its being developed by the French firm Sanofi-Synthelabo.

Summary:

A single pill has been found to stop both food and cigarette cravings enough to help people quit smoking and lose weight, and could also help curb other unhealthy urges.
Named Rimonabant Acomplia the drug in one study helped people who were overweight drop an average of 20 pounds and in another doubled the chances that smokers would quit.
Rimonabant (which will be brought to market as Rimonabant Acomplia) works by blocking the CB1 receptor, one of two receptors found in a newly described physiological system called the Endocannabinoid System (EC System), believed to play a critical role in the regulation of food intake and energy expenditure.
Researchers wondered whether a drug that halted this action might curb appetite, and in 2001, the first animal study was conducted at the National Institute of Alcohol Abuse and Alcoholism in Bethesda, Md.
When the normal mice were given rimonabant, which blocked their CB1 receptors, the mice reduced their food intake. In 2002, Sanofi-Synthelabo began human testing.
Rimonabant Acomplia works by selectively targeting and blocking the CB1 receptors, helping normalize the over-activation of the EC system and making hunger or cigarette cravings more manageable.
This blocking of signals that control cravings appears to result in weight loss, improvement of cardiovascular/ metabolic risk factors in overweight/obese patients and appears to reduce tobacco dependence without post cessation weight gain in people who smoke.
Meridia (Reductil in Europe) was originally a drug meant to treat depressions and can have severe side-effects as well, like high blood pressure, sleeping problems (Insomnia), constipation and dry-mouth syndrome.
This causes a lot of strain on many of the body systems and phentermine should therefore not be used for longer periods and only in obesity cases and not for treating common overweight.

Here’s another article that promotes the hype and mythology of Acomplia, an upcoming weight loss drug that still remains entirely unproven. Amazingly, even the fitness industry seems concerned about Acomplia now. But it’s ridiculous: no pill can make a person look fit. Nor can it actually improve a person’s cardiovascular health. There’s no replacement for physical exercise, but of course, Americans will probably buy into Acomplia anyway, because even the promise of weight loss without effort is good enough to cause an explosion in sales.
A much better approach to losing weight with the help of drugs is probably the upcoming PYY nasal spray product from Nastech / Merck. This uses a natural hormone (PYY) that you inhale to make you feel full more quickly.

Summary:

One of the big health stories of 2004 was development of Acomplia, also known as Rimonabant, a diet drug which shows real promise for sustained weight loss.
In early clinical trials, Acomplia is looking promising.
But with only 3,000+ people tested so far, it has a way to go before being approved by the FDA.
With the raging obesity problem, more and more people are seeking ways to lose weight without the pain and drudgery of dieting or working out.
The fitness industry frowns upon the notion of popping a pill to lose weight after all the battle cry is “The pain is the gain”.
Yet the idea of a diet pill is seductive.
It’s something that doesn’t require any exertion whatsoever, and that makes it a perfect fit for many Americans, who seem increasingly unwilling to actually take any action in order to achieve lasting results.
What they want is a magic weight loss pill.
And so far, Acomplia looks like the closest solution.
The drug’s manufacturer, Sanofi-Aventis, an international pharmaceutical, headquartered in Paris, plans to apply for FDA approval in 2005.
A decision by the agency will then take up to 10 months.
Diet pills have been around for decades offering different attempts at suppressing appetite or speeding up the metabolism.
But Acomplia offers a new technology known as a cannabinoid type I (CB1) blocker, it’s the first of an entirely new class of drugs, which works by blocking a pleasure center in the brain.
From the standpoint of researchers, the drug is clearly a breakthrough.
Will the entire fitness industry be challenged one day by the discovery of a magic diet pill?
